Kentucky Route 743 is a south–north route running through parts of Warren and Edmonson Counties in south-central Kentucky. Traversing farmland and woodlands for most of its length, it is primarily a rural access corridor allowing access to numerous higher numbered state highways.
Route 743 begins at a junction with US 31W in northeastern Warren County. It proceeds north for 1.5 miles before coming to a junction with KY 2630. The route continues on for another 2.5 miles before entering southwest Edmonson County.
Shortly after entering Edmonson County, KY 743 has a junction with KY 1749. The route then winds in a northeastern direction for another 3.6 miles before coming to a junction with KY 2326.
Route 743 then proceeds east for another 1.3 miles before coming to a junction with KY 101 in Chalybeate. Following this junction, the route has a concurrence with KY 101, heading south for 0.6 miles before splitting off to the east.
The route continues on for another 3.2 miles before coming to its northern terminus at a junction with KY 422 near Pig.
